Pamidronate Benefits Limited in Children with Osteogenesis Imperfecta

Source –Medscaoe – An article published in The Journal of Clinical Endocrinology and Metabolism 2006; 91:511-516 reports that in children with osteogenesis imperfect, the value of pamidronate therapy appears to be restricted to the first few years of treatment.

The researchers investigated the bone tissue effects of pamidronate therapy continued for an average treatment period of 5.5 years in 25 children and adolescents with osteogenesis imperfecta.

Average area bone marrow density increased 72% and volumetric bone marrow density increased 45% in the first half of the observation period, the authors report. In the second half of the observation period, however, they increased by only 24% and 14%, respectively.

The author suggests that “Preventive use of bisphosphonates in pediatrics is only justified in well-controlled studies”

Pamidronate is not licensed for use in children in the United Kingdom.
